Отца своего не знала и не видела, а её мать госпитализировали с психическим расстройством. Она перешла под опеку бабушки и дедушки, которые её вырастили и воспитали на ферме в Висконсине.
Обучалась в Университете штата Иллинойс. Там же она поступила в драматическую школу Гудмена, также выступала там на театральной сцене.
В 1963 году Фостер переехала в Нью-Йорк, где она продолжила свои выступления уже на Бродвейской сцене.
Успешно сыграла свою роль в пьесе «В белой Америке». Пьеса рассказывает об истории угнетения афроамериканцев в США. За свою роль Глория получила премию «Obie».
Сниматься в большом кино начала с 1964 года. Дебютировала в фильме «Параллельный мир», где сыграла роль Миссис Кастис.
Затем сыграла небольшие роли в «Я — шпион», «Комедианты», «Ангел Левин», «Мужчина и мальчик», «Шоу Косби», «Убийства детей в Атланте», «Закон и порядок», «Город надежды» и др.
В 1999 году исполнила роль Пифии — в популярном фильме «Матрица». Боевик рассказывает о хакере Нео, который испытывает скептицизм относительно истинной природы реальности, он знакомится с красавицей Тринити и ее шефом — дзен-хакером Морфеусом, который приглашает Нео совершить путешествие, открывающее разум. Нео узнает, что мир, где он раньше существовал — лишь компьютерная виртуальная реальность, управляемая машиной с искусственным интеллектом.
